The 2009–10 season was the 107th season of the Southern Football League, a football competition in the South and Midlands of England for semi-professional and amateur clubs. The season also saw Division One Midlands renamed Division One Central.

Three divisions competed that season: the Premier Division, Division One Central, and Division One South & West.

Premier Division
- The Premier Division had 22 clubs. Farnborough won the league and earned promotion to the Conference South.
- Nuneaton Town finished second and went through the playoffs to be promoted to the Conference North.
- Clevedon Town and Rugby Town were relegated.
- Merthyr Tydfil were expelled from the league at the end of the season, liquidated, and reformed later as Merthyr Town several levels below the Southern League.
- Tiverton Town and Hemel Hempstead Town were reprieved from relegation because other clubs higher up the pyramid folded.
- Top scorer: Stefan Moore (Halesowen Town) – 35 goals.

Division One Central
- This division had 22 clubs. Beaconsfield SYCOB, Burnham, and Slough Town joined from Division One South & West. Biggleswade Town also joined after moving up from the Spartan South Midlands League, and Hitchin Town was relegated from the Premier Division.
- Bury Town won the division and were promoted to the Isthmian League Premier Division.
- Chesham United won the playoff and were promoted back to the Southern League Premier Division.
- Aylesbury United were relegated; Rothwell United resigned from the league; Barton Rovers were reprieved from relegation.
- Top scorer: Jamie Arlick (Hitchin Town) – 29 goals.

Division One South & West
- This division also had 22 clubs. Windsor & Eton won the division and were promoted to the Premier Division; Cirencester Town also earned promotion via the playoffs.
- Bracknell Town were relegated.
- Top scorer: Justin Bennett (Gosport Borough) – 32 goals.
